Our technology is designed to treat diseased kidneys using a patient's own cells and may prevent or delay dialysis or transplant.ProKidney's Bioprocess R&D, Morrisville, NC is seeking a talented, motivated and skilled cell biologist/ physiologist as Principal Scientist (on-site) for studies relating to mechanism(s) of action of a cell-based therapeutic and its potency assurance strategy. The individual will be responsible for, and bring proficiency with, performing assays using 2D and 3D cell cultures including preparation of organoids and their biochemical and functional profiling. They will design experiments, record and interpret data, maintain equipment, and assist in ordering lab supplies. The individual must be detail-oriented, have strong communication skills, be conversant with the current literature and be willing to work in a dynamic, fast-paced environment. Independent reading and understanding of the scientific and technical literature for new assay development and troubleshooting is highly desired.ResponsibilitiesPerform tissue culture techniques to establish ex vivo kidney model systems from human samples (e.g. organoids in matrigel)Participate in the development of sophisticated in vitro model systems such as organoids and combination culture (assembloids)Utilize common molecular and cell biology skills including immunohistochemistry, immunofluorescence, single cell sequencing, etc., to characterize organoidsRecord all data and summarize in an organized fashionWork with the lab head and other members to design, develop, and execute experimentsRequirementsEducation/TrainingMaster's degree with 3+ years of relevant experience or PhD in cell biology/physiology or closely related field. Evidence of peer-reviewed published experience preferred.Experience4+ years of cell-based research experience.Skills/AbilitiesUse aseptic cell culture techniques to cultivate and expand human or other mammalian cellsPrepare 3-dimensional cellular constructs including organoids and assembloidsConduct cell function assays - cargo uptake including proteins and ions; ion exchange; tubule formation assay; organoid size and cell countSupport mRNA synthesis efforts from cell cultures and organoidsFluorescent immunoantibody staining of cells and/or organoidsImage capture and analysisProKidney is an Equal Opportunity Employer.
